Discovery could increase efficacy of promising cystic fibrosis drug

Mar. 21, 2013 ? A little more than a year after the FDA approved Kalydeco (Vx-770), the first drug of its kind to treat the underlying cause of cystic fibrosis, University of Missouri researchers believe they have found exactly how this drug works and how to improve its effectiveness in the future. Described in the current issue of the Proceedings of the National Academy of Sciences, MU researchers have redefined a key regulatory process in the defective protein responsible for cystic fibrosis that could change the way scientists approach the lethal genetic disease.

"They know the drug works, but they don't know how it works or where it works," said Tzyh-Chang Hwang, PhD, PNAS corresponding author and professor of medical pharmacology and physiology at the MU School of Medicine. "Our paper provides a theory for how Vx-770 works, and based on our understanding of how the CFTR channel works, we have identified a novel strategy for future explorations to complement and enhance the performance of the existing drug."

Cystic fibrosis is the second most common life-shortening inherited disorder occurring in childhood in the United States, after sickle cell anemia. Approximately 30,000 Americans have cystic fibrosis, and there are an estimated 1,000 new cases diagnosed each year. Cystic fibrosis patients are born with a genetic defect that causes a malfunction of the cystic fibrosis transmembrane conductance regulator (CFTR) protein, a chloride channel in the cell membrane that plays a critical role in maintaining water and salt balance across many body tissues, such as sweat glands, tissues that line the lungs, liver, pancreas and reproductive organs.

"The chloride channel is like a pipe that allows ions to travel through at a very fast pace," Hwang said. "In cystic fibrosis patients the channel is dysfunctional and activity is diminished. So what is the mechanism that controls the opening and closing of the channel? That is the fundamental discovery of our recent papers summarized in Physiology."

Like an automatic water faucet with a defective hand sensor, many genetic mutations found in cystic fibrosis patients cause a faulty signal, resulting in limited chloride transport across the CFTR. The result is the formation of thick mucus that builds up in the lungs, digestive tract and other parts of the body, which leads to severe respiratory and digestive problems, as well as infections and diabetes.

As summarized in Physiology and followed up with further research in the PNAS article, the accidental discovery of a mutation in CFTR, the R532 mutation, allowed MU researchers to reveal a new "non-strict coupling" relationship that occurs between the consumption of ATP, a molecule that provides energy in the body, and the opening and closing of the CFTR. They argue that the new information uncovered about this mechanism that controls the opening and closing of the CFTR and the passage of ions through it could explain how and where the new cystic fibrosis treatment Kalydeco (Vx-770) works.

"To his credit, Dr. Hwang exploited the behavior of the CFTR mutants to demonstrate that CFTR's gate is not strictly coupled to the nucleotide binding engine (NBD) that binds and splits ATP [energy] to drive conformational changes that regulate chloride flow through the CFTR protein channel," said colleague David Sheppard, PhD, an associate professor in the School of Physiology and Pharmacology at the University of Bristol in Bristol, U.K. who did not participate in the study.

In their study, MU researchers were able to observe the effects of the cystic fibrosis drug Vx-770 on the recently discovered R352 mutation. They found that Vx-770 enhances the activity of the CFTR channel by exploiting this "non-coupling" mechanism, a conclusion also supported by experimental results with the wild-type CFTR protein.

"Traditionally, researchers have defined how energy is utilized and transferred in the CFTR as a 'strict coupling' mechanism, meaning that one ATP molecule opens CFTR's gate, ions pass through and the ATP molecule is hydrolyzed and then the gate closes," Hwang said. "In this new model, we argue that the CFTR uses energy from ATP hydrolysis to carry out its function of chloride flow, but this coupling mechanism is more plastic than we thought and therefore could be subjective to manipulations by drugs such as Vx-770."

CFTR is part of a family of thousands of active transporter proteins called ABC proteins. Although CFTR may share many structural features with its ABC "cousins," as Hwang calls them, it has been unclear as to whether CFTR and its cousins may work in a similar manner.

The new idea of how the CFTR utilizes ATP to carry out its function may bear a broader implication because of the evolutionary relationship between CFTR and other ABC transporter proteins. It opens up a wide variety of therapeutic possibilities for other common diseases, such as cancer, heart disease and diabetes, Hwang said, since many other ABC proteins play critical roles in those human illnesses.

"It's taken years for scientists to solve this particular puzzle about the CFTR protein," Hwang said. "Our recent study provides evidence that these ABC transporter proteins and CFTR, a chloride channel, are two peas in a pod. Mother nature employs the same structural framework with just a little bit of modification to do two totally different things. From a basic science perspective, it's a big deal."

Using electrophysiology techniques available at MU's Dalton Cardiovascular Research Center, Hwang's lab studied the opening and closing, or "gating," of the CFTR at the single-molecule level. By measuring the electrical current that reflects directly the movement of chloride ions through one single CFTR channel as it opens and closes, Hwang's lab is able to monitor the activity of a single CFTR molecule in real time.

"Single-channel recording provides a unique opportunity to observe conformational changes in a single CFTR molecule in real time," Sheppard said. "It's exciting to think about how the new models proposed by Dr. Hwang and his colleagues explain the action of Vx-770 and other transformational drugs that target the root cause of cystic fibrosis."

Speaking at Your Local Library | ACFW Blog

Last November, my friend mentioned her book club was reading my first novel, Lakeside Reunion, and she asked if I?d like to come and talk at their November meeting. I agreed and enjoyed the conversation I had with them.

One of the book club attendees works as the business manager at my local library and asked if I?d be interested in speaking at our library. I agreed. She passed my name to the adult services librarian who schedules guest authors. The adult services librarian contacted me, and we scheduled a date for March.

While working out the details of my speaking engagement with the librarian, I learned a few things about speaking at libraries:

1. Be Prepared. During our email exchange, I asked questions about the library?s expectations for guest speakers-expected number of participants, any necessary equipment to be needed, time allotted for speaking, selling of books. By the time my speaking date arrived, I felt confident.

2. Be Yourself. I had asked if I should speak about anything in particular. She said, ?Be yourself. Readers love to learn more about the writer behind the stories.? I opened my talk with ?Everyone has a story to tell, and mine begins with a promise.? Then I shared about my writing journey and the way God?s promises allowed a small town girl?s dreams to become reality. Judging by their body language and eye contact, I assumed the guests were engaged in my talk. When I finished, several asked questions about different aspects of the story creation process and where my writing path was taking me now.

3. Be Passionate. I?m a card-carrying member of the Weepy Women?s Club. I cry over happy moments, tissue commercials and sharing my heart. During different moments of my talk, tears flooded my eyes, especially as I shared why Lakeside Reunion was the story of my heart, getting ?the call? from my agent and other exciting writing achievements. I made no apologies because that?s a part of who I am. I was able to share my faith and my love for Christian fiction without being preachy. And I saw a tear or two in the audience.

4. Be Generous. At the conclusion of my talk, I gave away two gift baskets containing coffee, tea, cocoa, mugs, chocolate and CBA novels. I donated two large print copies of my first two books-Lakeside Reunion and Lakeside Family-to the library. Also, I made a donation to our Friends of the Library fund to help purchase more books. All of these contributions cost me little, and they can be deducted as business expenses on my taxes. Your contributions to your library are remembered, especially when readers check out your autographed novels and see Local Author stickers on the spines.

5. Be Thankful. I thanked the librarian for inviting me, and I thanked the guests for coming to join me. I followed up by sending handwritten thank you notes to the library staff who helped make my first speaking engagement at the library a success.

Speaking at your local library is a great way to build community relationships and connect with your local readers.

Subway worker stuck in chest high mud 75 feet below ground

NEW YORK (AP) ? A subway construction worker was stuck in mud for hours in an underground tunnel in New York City before being rescued.

Fire officials say the worker was pulled out shortly after 12:30 a.m., about four hours after he became stuck up to his chest in the tunnel 75 feet below ground. Authorities say he was treated for hypothermia and is in serious but stable condition at a hospital.

More than 150 firefighters responded to the site on Manhattan's Upper East Side, where the Second Avenue subway line is under construction. Three firefighters have suffered non-life threatening injuries. A Consolidated Edison vacuum truck helped remove debris.

It happened at around 8:30 p.m. Tuesday.

In September, a controlled blast at the subway construction site sent rocks flying into the streets.

We're here at NVIDIA's GPU Technology Conference in San Jose, and company CEO Jen Hsun-Huang has just revealed the next step in its GPU roadmap. Called Volta, it's scheduled to arrive after Maxwell, and will advance GPU technology with a ridiculous amount of memory bandwidth. Volta GPUs will have access to up to 1TB per second of bandwidth by stacking the DRAM on top of the GPU itself, with a silica substrate between them. Then, by cutting a hole through the silicon and connecting each layer it's possible to move, according to Huang "all of the data from a full Blu-Ray disc through the chip in 1/50th of a second." We aren't exactly sure what that means for graphics, but being able to process data that quickly is bound to be a boon for gamers... whenever Volta actually arrives, of course.

US reassures Poland over missile defense changes

WARSAW, Poland (AP) ? U.S. officials visiting Warsaw tried to reassure Poland on Monday that the cancellation of the final stage of a European missile defense system will not sideline the country and was not made to appease Russia.

U.S. Defense Secretary Chuck Hagel announced last week that plans to place long-range missile interceptors in Poland in the next decade are being abandoned and that interceptors will be placed in Alaska instead due to the threat from North Korea.

But U.S. defense and political officials on a previously scheduled visit to the Polish capital said shorter-range interceptors would still be based in Poland as part of NATO's missile defense system in Europe.

The Obama administration has planned to put medium-range interceptors in Redzikowo, in northern Poland, in 2018, as part of the NATO system. They are meant to protect Europe from attacks from Iran.

Wendy Sherman, U.S. undersecretary of state for political affairs, said the Polish site "will go forward as scheduled."

The U.S. missile defense plans involve building up the system in Europe in four stages, with shorter- and medium-range interceptors to be deployed in the first three phases, and longer-range interceptors due in the fourth phase.

However, the fourth stage has not yet been funded by Congress and there are indications the technology is not ready. At the same time they have been an irritant in relations between the U.S. and Russia, which suspects the interceptors were meant as a counter to Russian missiles ? something Washington firmly denies.

Nonetheless, Moscow had indicated that it would not consider further nuclear arms cuts unless its concerns were resolved.

Past efforts by President Barack Obama's administration to alter the missile defense program have sparked criticism in Poland. This time, however, Polish officials declined comment on the matter.

Poland's main aim in having the U.S. interceptors has been to have an American military presence on their soil in the belief it will increase their security, particularly given fears that Russia could one day try to dominate them again.

During a news conference in Warsaw along with Polish officials, Sherman was asked by a Polish reporter if the move was meant to mollify Russia.

In her answer, she noted that U.S. Secretary of State John Kerry told his Polish counterpart of the decision before announcing it publicly ? a courtesy not extended to Moscow.

"Russia was only notified after those calls," Sherman said.

Phase one of the system has already been deployed, with anti-missile interceptors on a ship in the Mediterranean Sea. Phase two is to include interceptors in Romania, then interceptors in Poland will be phase three.

"For Poland there is no change, because the phase-three system, which was always planned to be in Poland, is still now planned to be in Poland," Madelyn Creedon, the U.S. assistant defense secretary for global strategic affairs, told The Associated Press. "It's funded, it's committed. That was our original plan and we haven't changed it."

The U.S. missile defense system at Romania's Deveselu military base is not affected either and will become operational in 2015 as planned, the Romanian defense ministry said Monday.

No criminal charges for U.S. parents of adopted Russian boy who died

SAN ANTONIO (Reuters) - No criminal charges will be filed against the adoptive parents of a Russian-born boy who died in January in Texas, a prosecutor said on Monday, in a case that has intensified a dispute over international adoptions.

The January 21 death of Max Shatto, 3, sparked criminal and child welfare investigations in Texas, probes by authorities in Russia, and calls from Russian lawmakers for his younger brother to be returned to Russia.

Max was found dead in the yard of his home in Gardendale, Texas, near the oilfield city of Odessa.

A West Texas grand jury determined that there was no evidence that his adoptive parents, Laura and Alan Shatto, had committed any crime, Ector County District Attorney Bobby Bland said in a statement.

"This death was the result of a tragic accident occurring most likely on playground equipment," Bland said. "When a child dies so young and tragically, it is natural to want to hold someone accountable. However, in this case, there is no evidence to support holding anybody criminally responsible."

Oceanic whitetip sharks, porbeagle sharks, scalloped hammerheads, great hammerheads and smooth hammerheads ? as well as two species of manta rays ? are set to get new protections after this week's votes at the meeting of the Convention on International Trade in Endangered Species of Wild Fauna and Flora (CITES) in Bangkok.

If the proposals are upheld at a plenary session later this week, all seven animals will be listed under Appendix II of the CITES Treaty, which includes species that may become threatened with extinction if they are traded unsustainably. So far, basking sharks and?great white sharks?are the only species of elasmobranch (a family that includes sharks, rays and skates) listed on Appendix II.

Sharks are apex predators that?help balance ecosystems?in the world's oceans, and they have slow growth and reproductive rates, making it difficult for their populations to bounce back from big losses. The votes at CITES were applauded by conservationists and biologists who say overfishing is by far the biggest pressure faced by sharks. [On the Brink: A Gallery of Wild Sharks]

The fish are harvested for their meat, liver oil and cartilage, as well as their fins, which are cut off to be used in?shark fin soup, an ancient and prized delicacy in East Asia. According to the World Wildlife Fund, a shark's fin can sell for up to $135/kg in Hong Kong.

"That market has created a lot of demand for shark fins and even spawned a brutal practice in some fishing communities called 'finning,' in which sharks have their fins cut off (the most valuable part) and are then thrown back alive but finless, where they most certainly die,"?Alistair Dove, a marine scientist at the?Georgia Aquarium, told LiveScience in an email."Manta rays are facing a similar challenge, except that in those species it is the gill rakers that have developed a market for use in Chinese traditional medicine, leading to unsustainable harvest of those peaceful and graceful plankton feeders."

David Shiffman, a shark biologist, told LiveScience that for the species what will be covered under the new CITES listing, "a lot of the trade is largely unregulated and it's led to massive population declines, particularly for hammerheads and oceanic whitetips."

Though the decline of shark populations has been documented, scientists don't have great data on how bad the problem is since unregulated and illegal catches often go unreported. A study out earlier this month estimated that about?100 million sharks are killed?each year, but researchers said the real number of annual shark deaths could fall in a rather large range, between 63 million and 273 million. Shiffman was hopeful that more regulation will give scientists a better idea of the numbers.

"This will lead to additional data about the harvest of these species, which will allow us to make more informed management decisions in the future," Shiffman said. "More data is always better for science."

To help drum up support for shark-protecting proposals, the U.S. delegation to CITES says it worked with other countries, including Brazil, Colombia, the European Union, Costa Rica, Honduras, Ecuador, Mexico and Egypt.

"We are extremely pleased that CITES member nations have given greater protections to these commercially exploited marine species," Bryan Arroyo, head of the U.S. delegation, said in a statement. "Through the cooperation of the global community, we can begin addressing the threats posed by unsustainable global trade in shark fins and other parts and products of shark and ray species."

The CITES Treaty is signed by 178 countries, and a meeting is held every two to three years to review and negotiate changes to the international trade of species covered by the agreement. Whereas sharks fared well during this year's meeting, another highly anticipated proposal to?ban the trade of polar bear?hides and parts was shot down.

(AP) ? Bayern Munich barely staved off an improbable comeback attempt by Arsenal and advanced in the Champions League despite a 2-0 loss Wednesday night, leaving England without a team in the quarterfinals for the first time in 17 years.

While the Gunners' win tied the home-and-home series 3-3, and Bayern Munich advanced because of a 3-2 advantage in away goals.

After losing the first leg 3-1 at home on Feb. 19, Arsenal went ahead on Olivier Giroud's goal in the third minute. Laurent Koscielny scored in the 86th to tie the aggregate, but the Gunners never threatened again.

"Our second goal came a bit too late," Arsenal manager Arsene Wenger said. "Bayern was nervous, but we paid the price for not playing well in the first leg."

Bayern, a four-time winner and runner-up twice in the last three seasons, joined Barcelona, Real Madrid, Malaga, Juventus, Borussia Dortmund, Paris Saint-Germain and Galatasaray in Friday's quarterfinal draw.

"It shouldn't have been that close," Bayern forward Arjen Robben said. "We talked about it before the match. Arsenal had nothing to lose, and we were not aggressive enough. Maybe it was a wakeup call. We conceded an early goal and there were still eight minutes to play after their second, we knew we were close to letting it slip away."

Malaga became the third Spanish team to advance, beating visiting Porto 2-0 for a 2-1 aggregate win as 20-year-old Francisco Alarcon scored in the 43rd minute and Roque Santa Cruz in the 77the.

Porto's Steven Defour was ejected after receiving a second yellow card for a late challenge on Joaquin Sanchez in the 49th minute.

Germany has two teams in the final eight for the first time in 11 years.

Tie the string at the top, and you?ve got a bird feeder. Have your kids use biodegradable non-toxic paint to decorate the bird feeder, and hang it in your yard (filled with seed of course)!

Pencil Holder
Your kids will love making a recycled pencil holder that they can take to school to show off to all of their friends. Take some old, small milk cartons and wash and dry them. With scissors, cut off the top of the milk cartons so you have a tall, square box. Using wallpaper or construction paper, line the inside and outside by gluing the paper to the carton.

You can let your children pick out the colors of construction paper or wallpaper, and then color them however they?d like. It?s a great way to recycle milk cartons, and it?s a great activity for kids at a birthday party so they have souvenirs.

Leaf Prints
Another great idea for a green arts and crafts projects for you and your children to do is creating leaf prints. Have your kids go outside and collect a few of their favorite leaves, and then spread out some newspaper onto the table or floor. Take a single leaf and flip it to the side so its veins are raised. Then have your child paint the leaf, and press the painted side onto some white paper. Then, all you need to do is flatten the leaf and lift it off to reveal a beautiful print.

You can use the same leaf multiple times, or opt to make a collage with multiple leaves. You can use it as an educational opportunity to teach your children about the different kinds of plants and trees in your backyard!

(AP) ? The dead vulture's feathers snap and crack, breaking apart as its frozen wings are spread for one last flight.

It will soon soar gracefully ? albeit briefly ? into a tree in this hilly New Jersey suburb, hoisted to a branch where it will hang, upside down, until spring.

Wildlife officials say it's a sure-fire way to get an estimated 100 black and turkey vultures from roosting in the neighborhood, leaving behind foul-smelling and acidic droppings on roofs and lawns, creeping out residents and even their pets.

Before the black vulture's carcass is strung up, nearly a dozen vultures glide over Bridgewater on a cool, gray Monday morning. Some perch in trees. One rests on a chimney-top.

Neighborhood residents watched as wildlife specialist Terri Ombrello launched a weighted fishing line over a branch with a sling shot. She took turns with partner Nicole Rein tying the bird's legs with another line then pulled the bird about 30 feet off the ground.

Vultures may like to eat road kill but it turns out they don't like the sight of their own dead upside down.

"They don't like seeing their own in that unnatural position," Rein said.

Bridgewater, a town of 45,000 about 40 miles west of New York, became at least the seventh New Jersey community this winter to turn to the wildlife services unit of the U.S. Department of Agriculture for buzzard-beating help. Black and red turkey vultures are protected species and cannot be killed without a permit.

The birds roost from November to April, settling down as it gets dark, when they are most visible.

Jim Van Allen, 69, lives across the street where the carcass was strung up in Bridgewater. He's lived in the neighborhood practically his entire life. He said it isn't unusual to see vultures there in this community but not this many. He said the vultures started arriving in November, just after Superstorm Sandy.

"They just glide all around, all day long, I mean, just looking for something dead," he said.

The vultures, which have sometimes lined up eerily on rooftops, have not just spooked residents. Mark Nathan said his yellow lab Callie is afraid of the vultures, especially when they fly low.

"She freaks out about them," Nathan said. The dog "barks at them and then she runs inside as fast as possible," he said.

Scavenging vultures are key to the ecosystem because they feed off dead animals, acting as flying garbage disposals. Still, in densely populated areas where they can thrive, vultures pose a serious nuisance.

"Their feces runs down the roof. It looks bad," Van Allen said.

Residents can expect to see fewer vultures within one to three days. While some may still perch on the tree, Rein said, they will not do so for long.

While some New Jersey towns regard the influx of vultures as a problem at least one community is hoping it will get its birds back.

Wenonah started holding a vulture festival in 2006 after nearly 200 turkey and black vultures made the town their winter home. But the town canceled its festival this year, according to Vulture Festival website, because they're no longer roosting there at night.

'Dating' Other Mothers - NYTimes.com

?I have to start dating again!? I wailed to my husband. It?s probably closer to the truth to say that my 3-year-old and I were going to have to start dating again.

At this moment, one of my closest local mom friends, Katie, is packing up and readying her family to move to Wisconsin. Katie and I go on weekly walks together, cart our toddlers to music class and park dates, and fit in what other socializing we can. With Katie, I could always count on a sympathetic and nonjudgmental ear about all things. But now she?s leaving me for the Midwest. To make matters worse, Kristen, the other mom friend I completely adore and have been fast friends with since pregnancy, is also moving out of state.

In Katie and Kristen, I found the sort of kinship in which I felt comfortable expressing any and all concerns or stresses about my son and motherhood. With the busy schedules we each maintain on not enough hours in the day, finding moms you really click with on a parenting as well as a friendship level can be fairly difficult. Maybe you like each other, but your kids don?t get along. Or maybe your kids are great friends, but you have nothing in common with her. Or your kids are different ages, which matters much more than I ever would have thought.

Neither Katie nor Kristen has even left me yet, but I?m already mourning their loss and desperately casting about for replacements. I employed this strategy with crushes throughout high school. There would be my primary crush, but there would also be two or three backup crushes so that in the event of a breakup, I?d instantly have another romantic target to daydream about and take the sting out of being dumped.

A few weeks later, my son and I were driving off for a ?first date? with another mom and her two girls. Our kids had hit it off so well in a new music class that at the end of one session, I fingered my business cards in my purse. I was all primed to give her my e-mail address at what I judged to be precisely the right moment. I was going for breezy and relaxed, not desperate and needy. Impatient, Bug grabbed a fistful of my cards, and trotted over to the other mom with them. There?s nothing like a toddler for timing.

Rushing around the house to get ready for our first date at a park, I felt so buoyed by the adrenaline of anticipation that I had to dump out the rest of my coffee. No need to be that jittery and motor-mouthed on this first outing.

Deciding what to wear was of paramount importance, of course, and I will admit to trying on several outfits before being satisfied. I was going for a look that wasn?t too cute or too dressed up (because impeccably put-together moms are off-putting), but also avoided boring or sloppy. My makeup application followed a similar line of thinking ? restrained and natural, but enough to keep the haggards at bay.

I wondered if our age gap would end up being a problem, compatibility-wise. Seven years my junior, she is just one year older than my baby sister. When she graduated from college, I had already been married for over two years. But I?ve since realized that when you have kids, it?s no longer your age that matters. Who cares if you didn?t both watch ?90210? in high school when you can both Netflix it now? Far more significant is the age of your kids and whether or not they are in sync with one another, because that alone gives you a lot of common ground and endless conversation fodder. It?s a lucky bonus if you discover you also have other shared interests. In fact, with her two kids, my new mom-friend is older than me in terms of motherhood experience. Especially since her first girl is six months older than my son, which seems to give her a wise window into what I can expect down the rocky toddler road.

As it turned out, the chatter never lagged, and we had a fabulous time at the park. The kids played amazingly well together, giving us time to talk and find out all sorts of things we had in common. Books, television and our obsessive love of our pets and of foggy, dark days were significant points of interest. (Later, I would learn that she also loved Ioan Gruffudd, tawny port, ?Friday Night Lights,? ?Hart of Dixie? and other things that would prompt me to call her my doppelg?nger.) The date was a huge success ? right up until my son took his first major spill on a playground and got a mouthful of wood chips and sand. He tried to rally during snacks, but it was clear that his sunny mood was killed by the slightly split lip. Poor kid.

That night, I got a sweet e-mail from her checking in on how my son was doing and sending hugs from her and her girls. It was the equivalent of a postdate ?I?m still thinking about you? check-in. In my response, I told her that despite his painful fall ending his park play, Bug announced, ?I love Lila,? during the car ride home. This was significant, not only because it was the first time he had said he loved a nonfamily member, but also because it?s just been recent days that he?s been saying it voluntarily at all.

?You played the love card too early!? my husband teased me the next day when I didn?t hear back from her. Lord help me, but I had the same thought! Did I come on too strong? Was I a bit too keen? By now I was calling her Mom Crush on Twitter and tweeting ecstatically about our new friendship.

The next music class arrived, and as soon as her littlest saw my son come in the door, she toddled up and embraced him solemnly. My son was slightly surprised, but then he bent his head to her tiny 17-month-old shoulder and hugged her back. Clearly, there was enough smitten to go around, so I told her that my husband had joked about an arranged marriage between the kids.

?? And THEN she started quoting Jane Austen from the Lady Catherine de Bourgh scene, Mom!? I told my mom on the phone that night. ?Oh, she sounds just lovely, Honey!? my mom said. That?s right, I called my mother to tell her about my new mom-friend.

Meanwhile, Mom Crush and I went on second and third dates with our kids, and have now exchanged books and many e-mails, and set standing appointments to go on early-morning weekend walks without the kids.

I?m considering making her a mix tape.

Bronze-Age Donkey Sacrifice Found in Israel

Archaeologists in southern Israel say they've uncovered a young donkey that was carefully laid to rest on its side more than 3,500 years ago, complete with a copper bridle bit in its mouth and saddle bags on its back.

Its accessories ? and the lack of butchery marks on its bones ? lead researchers to believe the venerated pack animal was sacrificed and buried as part of a Bronze Age ritual.

Donkeys were valuable beasts of burden in the ancient Near East. Donkey caravans helped open up vast trade networks across the Levant and Anatolia in the 18th and 17th centuries B.C., according to archives from Amorite settlements like Mari in modern-day Syria. Ancient Egyptian inscriptions from around the same time show that hundreds of pack donkeys were used in large-scale expeditions to mining sites in the eastern desert and southern Sinai, researchers say.

The animals have even been associated with royalty. In 2003, paleoscientists discovered the skeletons of 10 donkeys nestled in three mud graves dating back 5,000 years ago when Egypt was just forming a state. The donkey skeletons were lying on their sides in graves at a burial complex of one of the first pharaohs at Abydos, Egypt. [See Images of the Sacred Donkey Burial]

The donkey found in Israel seems to have been symbolically important, too, though this particular animal likely was never made to do hard labor before its death, said a team headed by archaeologist Guy Bar-Oz, of Israel's University of Haifa.

The grave was found in a temple courtyard, in the heart of the sacred precinct of Tel Haror, a Middle Bronze Age city near Gaza that was fortified by massive ramparts and a deep moat and dates back from around 1700 B.C. to 1550 B.C.

The donkey, estimated to be about 4 years old, was laid on its left side with its limbs neatly bent, the researchers say, and a copper bridle bit, a mouthpiece used to help steer animals, was found in its mouth. Some parts of the bit were extensively worn and it likely wasn't functional at the time of the burial. But an examination of the donkey's teeth suggests it was never meant to be practical.

"The absence of any sign of bit wear on the lower premolars indicates that the animal was not ridden or driven with a bit for prolonged periods of time," the researchers write in a paper published online this week in the journal PLOS ONE. "Moreover, the young donkey was still in the process of shedding its teeth and permanent teeth were just erupting. The researchers say this is the only known Bronze Age bridle bit to be found in the mouth of an equid and that it likely served as a symbol of status, evoking the chariots that pulled soldiers, people of high-rank, and in a ritual context, images or statues of deities.

There were no butchery marks or burning traces on the donkey's bones, suggesting the animal was not killed to be eaten. In contrast, a pile of scratched-up bones from sheep and goat were discovered just above the donkey's carcass, which the researchers believe could be evidence of a feast after the ritual slaughter.

AT&T to start selling BlackBerry Z10 devices on March 22

TORONTO (Reuters) - AT&T Inc will start selling the highly anticipated BlackBerry Z10 touchscreen smartphone in the United States on March 22, the company said on Monday, sending BlackBerry shares more than 10 percent higher.

BlackBerry , formerly known as Research In Motion, is hoping the new devices, already on sale in Canada, Britain and more than 20 other countries, will help it win back market share in the United States, which was once a stronghold for the smartphone industry pioneer.

The U.S. launch of the new devices has been delayed due to a longer carrier-testing phase in the country. AT&T said pre-sales of the devices will begin on Tuesday, March 12.

BlackBerry says sales of its new smartphone have been outpacing its expectations so far, but investors are keen to see how it fares in the United States. The company has lost U.S. market share rapidly to Apple Inc's iPhone and a slew of devices powered by Google's Android operating system.

As expected, AT&T said it would sell the devices for $199.99 with a two-year contract. T-Mobile USA said on Friday it planned to start selling the BlackBerry Z10 to its business customers in the United States on Monday.

Verizon Inc , the biggest U.S. wireless carrier, has yet to say when it will start selling the Z10. The Z10 and the soon to be launched Q10, which will come with BlackBerry's traditional physical keyboard, are powered by the new BlackBerry 10, or BB10 operating system.

"We believe the Street is pricing in such a weak fiscal 2014 that BB10 does not need to be an outstanding success to surprise," Scotiabank analyst Gus Papageorgiou said in a note to clients on Monday.

Papageorgiou, who has a "sector outperform" rating on the stock, said he expects the company sold about 1 million BlackBerry 10 devices in the quarter ended March 2.

"Gross margins should begin to move higher as more Z10s enter the mix," he said. "Next quarter will be the true test as BB10 launches in the U.S."

BlackBerry shares were up more than 10 percent at $14.37 in mid-morning Nasdaq trading on Monday, while its Toronto-listed stock rose by a similar margin to C$14.78.

Heads up, Windows users. Tomorrow, Microsoft will release an update for Internet Explorer 10 that enables Flash content in both Windows 8 and Windows RT. As many of you are likely aware, the "full web" experience has been limited to the desktop browser on Windows 8 up until this point, which was an intentional move by Microsoft in order to improve performance, battery life and the touch experience. With the update, Internet Explorer 10 users for Windows 8 / RT will be able to access Flash content on all but a few sites that Microsoft has selectively blacklisted due to their negative impact on the user experience. Naturally, users of IE10 within the Windows 8 desktop environment will still be able to access all Flash-enabled content, regardless of whether the site is on the blackballed list.
